The Process of Protein Purification Literature review

The Process of Protein Purification - Literature review ExampleVaccines, factor therapy and replacement (such as that in insulin-deficiency) all have helped in improving health conditions, and have been based on good elucidation of structures, research on structure-function relationships, and establishment of protein purification specific to the amino acid sequence present. With this in mind, this particular study designed a protocol to purify and characterize the a synthesis of cytochrome oxidase (SCO)-1-like protein 3966 in Streptomyces lividans. As will be seen later, better understanding of SCO proteins is still warranted, as many potential functions of these types of proteins are unclear. Moreover, SCO is a vital enzyme as the cytochrome oxidase c, and in essence the electron chain transport of the mitochondrial respiration mechanism, depends on it.Initial studies of homologues in bacteria have been the usual first step in protein characterization. Many proteins in the eukaryoti c cells have been proven to have functional and structural counterparts in bacterial cells. Because of the relative ease of bacterial replication and protein purification, it is gum olibanum a method of choice in conducting in depth studies of proteins.REVIEW OF RELATED LITERATURE I.Protein Purification at that place are factors to consider in doing protein purification. ... There are many kinds of column chromatography, ion-exchange, affinity, and size exclusion are just some of the more usual protein purification procedures that may be done. Affinity chromatography uses antibodies for a specific protein as part of the column through which the protein solution passes. Although it is highly specific, it is more expensive and much harder to prepare. Size exclusion, on the other hand, depends on the differences of molecular(a) regardts of the proteins that are present in the solution. In general, proteins with high molecular weight are eluted fastest as they are not able to get into the atrophied spaces of the column, making their path down the column less impeded. On the other hand, low molecular weight proteins still pass through the tiny spaces at bottom the column, thus slowing down their descent. Although much easier to prepare than an affinity column, a size exclusion chromatography column is less specific, as different proteins of similar weigh are eluted out at the same time, despite them having differences in characteristics, such as the isoelectric point (Burgess, 2008).. For ion exchange chromatography, these beads are charged, thus attracting the oppositely-charged proteins present in the solution to be passed through the column. Depending on the objective of the experiment, the eluent or the outflow proteins are collected for further processing such as concentrating. To get the proteins bound on the beads, salt solutions of graded concentrations are passed onto the column. As the concentration of the salt increases, the beads will more likely b ind to the salt than to the proteins. Thus, weak bean proteins are bound weakly

This physical journey is also an inner quest in search for immortality. The antics of an ancient woman are fully described and the likeness of old Phoenix grows clear to us. In just the first few lines, Welty has fully ingrained this old and wrinkly woman before us and presented us with a suspending question at the same date, why is this very old and small (Welty, 1941, paragraph 1) woman making a journey alone in the cold? Each one is different but the stones are exactly as she remembers and this is significant because in the scenes after, she seems shot of the way things should look. Like Welty, Rhys has presented us with a suspending question here, what has taken place that makes the protagonist reacts thus? The missions of the quests in both works are not made clear to the reader till the end of the story. However, both Welty and Rhys have used the setting extensively to show the protagonists progress with the journey, the passing of time and their state of mind. The characters of old Phoenix and the ghost girl are fleshed out so well that towards the end of the journey, not only are the quests explained, we also read with the characters. As old Phoenix embarks on her journey, Welty let us know that old Phoenix is familiar with the path but the journey is long. Alo ng the journey, old Phoenix has to cross different terrains. The pinewood at the onset of the journey marks the starting point of old Phoenixs inner quest for immortality, as pine trees are often associated with immortality. The quest is not without a struggle. It is an uphill battle but old Phoenix is resolute. She then has to make her way by dint of a forest of oak trees, oaks representing strength and wisdom to be gained. At this point, Welty has aptly

Early on the story, Charlotte Fielding (Winona Ryder) and Will Keane (Richard Gere) fell in love, and as Keane was ending the relationship in the same manner as he had ended his past relationships, Fielding agreed to the fact that they would have no future together because she was sick. The story ended in tragedy as Fielding dies of a rare disease after the last-resort surgical operation failed to save her life. There are legion(predicate) similarities in the cardinal movies in the manner that cancer was portrayed. First, the illness was equated to the certainty of death. The drama in both films revolves around the fact that the women have little date left. Both protagonists also dealt with the disease in a similar manner - they gave up on medical treatments. In Autumn of New York, Charlotte Fielding until now signed a directive that no surgery will be made on her. In one of their conversations, Fielding told K eane, I dont want to top hope, when there is none (Chen, 2000). The same attitude was somewhat projected by Sara Deever in Sweet November. She decided to take on a different menstruate in life, away from the regular treatments usually provided by hospitals to cancer patients. Both films in a way concluded that there were no other options for the two cancer-afflicted characters, and that subjecting themselves to medical treatments and procedures would yield the same result, that is, eventual death. The character in Autumn in New York died at the end of the film, while in Sweet November, the character decided to come home and perhaps yield to medical cure for her illness, although the last scene seemed to indicate that she would be spending the rest of her life wilt away and succumbing to death. She left the viewers thinking that she would die, in the same way that her lover agreed to letting her go and face death. Both stories depict the death characters, both of which were white women, still in flawless appearance. This would fall under what Comedienne Carol Burnett referred to as the Movie Star Disease (as cited in Lallanilla, 2005), where the ill character,

The understanding of modern urban life and the place of a domain in modern city - Essay ExampleIn this work Simmel underlined that in conditions of modern city and urban society an individual can successfully develop his skills and abilities, as the economical and political situation of modern and post-modern community promotes individual freedom and flexibility. Simmel underlined that the city conspires to erase difference by assaulting the individual with an overcome and never-ending stream of visual stimuli (Byram, 2002). This vision is inherent to the spacing theory of scientific rationalism, modern and post-modern experience that fills the space with giant cubic forms suppressing the consciousness of lot and their freedom. In Simmels opinion, our society consists of individuals who interact with each other, and this interacting forms features and specificity of our society While there is no perception of society until individuals begin to interact, once formed by the interac tion of individuals, the society affects the individuals as an outside force (Crow Ch., et al., 2000).Benjamin considered modern urban experience and a new politics of vision through the development of technologies. He underlined that because of mechanical reproduction, art loses its authenticity. Simmel also underlined that metropolis allows individuals to develop their freedom and art. But in the other hand, Simmel asserted that post-modern urban spirit threats individuality (see above). As salubrious as Simmel, Benjamin had positive attitude to discourses development Benjamin is positive about new technologies, emphasizing their liberating, democratising stoops. Simmel gave to a greater extent social approach to the communication technologies as interactions between people asserting that they form our society, but Benjamin wrote about the positive attitude to such technologies as film and photography crude media technologies such as phonographs, epic theatre, and especially f ilm and photography, not only destroy arts aura but demystifies the process of creating art, making available radical new glide path and roles for art in mass culture (Bicket, 2001). New World vs. Old Essay Example for Free

new-fashioned knowledge domain vs. Old EssayGlobal Wine War 2009 novel World versus Old 1. How were the French able to dominate the worldwide wine trade for centuries? What sources of competitive advantage did they develop to support their exports? The wine production of France goes all the manner back to being part of the first niche market for premium wine. Factor conditions of France, such as land and climate, were well suited for wine-production compared to other European countries.Since the early 19th century cross-border shipping was very expensive, France was able to cluster the related and supporting industries of wine production within their domestic borders. As a result, France increased its competitive advantage throughout other sources without much impact from other countries. As the industry became Frances second largest export, slope by side with a culture of rich food, the pauperization conditions got affected, as consumers were expecting a certain quality.T he French government provided the classification systems, which increased the rivalry and domestic competitiveness and raised the entry barriers so that entry of foreign competition was kept at a minimum. 2. Given the farseeingstanding dominance of Old World wine producers, how were the natural World producers, such as the Australians, able to expand their market share so rapidly in the 1990s?As postwar increased demand for wine rapidly, factor conditions of the New World producers, such as widely available suitable land and it being less expensive, made it possible to edge the new demand and boosted the New World industry. The new demand conditions demanded higher quality wines and as new generations were born, the consumers got to a greater extent price-conscious and convenient oriented. New World producers met these changes in demands by introducing a completely new firm strategy, structure, and rivalry compared to the Old World producers, who were constrained by tradition. In novation was the key word in the New World strategy suddenly, mechanical technology reduced labor cost and pushed down prices, packaging innovations made distribution easier and provided convenience to the consumers, and technology ensured vintage-to-vintage consistency and so forth As well, branding and marketing skills were performed on the basis of vintage-to-vintage consistency, which gave the consumers further preferences and strengthened the position of the New World producers globally. This made them capable at expanding even more rapidly. The New World producers changed the traditional patterns of related and supporting industries, which used to being long value-chains, as the big producers now controlled the full value chain, able to quote margins and control quality at every level. The size now gave them bargaining power a power, which made them have a strong grip on the importing countries. The New World producers had created a strong innovative company culture a culture that could easily adapt to the changes of consumer culture and government regulations.New generations were born and so was the rise of fashion in a completely different culture of lighter consumption. The New World producers responded to the swings of fashion by victorious advantage of the factor conditions much capacity and regulatory freedom. This was the complete opposite to what the Old World producers were able to do still constrained by the long history of wine-making tradition and lack of innovation. By that, the New World producers were always a few steps ahead of the Old World producers when the global patterns changed.

Mary Shelleys Frankenstein Essay Example for Free

Mary Shelleys Frankenstein EssayLater on the giant compares himself to exaltation Remember that I am thy creature I ought to be thy Adam. Just desire Adam the monster was created by god ( creating life ) which is ironic because this is the take in which people believe Viktor is trying to idolise himself to. In the society they lived in they were very religious and the fact that it werent god who created the monster this would be seen as sinful. It alike shows a moral meaning and relation to Did I request thee, Maker, from my clay To mould me man? Did I solicit thee From darkness to promote me? (John Milton, paradise lost). This source is asking did I ask to be made at the same time saying there evil-the maker, like what the monster is trying to get superordinate to see. Shelley chooses this metaphor to show that the monster wishes to belong his creator. The reader feels sympathy here because we get the impression that the monster is lost or unloved, longing for some reas on of family. The monsters explanation of his treatment by man All men hate the wretched how then, must I be hated, who am miserable beyond each living things. Here the monster mentions how he has suffered pain by man he believes Viktor wishes him to suffer further as a type of punishment. The reader sympathises with the monster due to his previous treatment by man, we believe it is Viktor who should be made to suffer. We get a real sense of the monsters loneliness when he asks Viktor to make him a female companion like him so he can be loved for once. At the end dramatic language makes the reader feel gloomful for the monster, I shall die and I shall no longer feel the agonies that consume me. This makes you feel sorry for him because hed rather die and has to pity himself because no one else does. furthermore the monsters feelings come across strongly when he says he is doomed.I shall collect my funeral pile and consume to ashes this miserable frame. This is so sad because he is planning his cause funeral Mary Shelley was trying to make people shouldnt judge by looks its the person inwardly that counts. She was also trying to suggest that people cant be born(p) evil its how they are nurtured which develops their personality I think Mary did get the message across quite clearly. She first came around about(predicate) writing the fabrication in a villa in the setting Swiss mountains and the lakes where there was always a stimulating conversation.Lord Byron suggested they all tell ghost stories to one other as an added incentive they decided to turn into a competition to see who could come up with the scariest novel. But Mary went on to develop her short description she came up with that night into a full novel which got published in (her version) in 1823. Like the people she was surrounded by Mary had a feature keen interest in science which is what her novel is based around. Darwin was a respected poet and scientist amongst Marys family and when he studied a piece of vermicelli within a glaze jar make voluntary actions of its own. This triggered the thought that corpses could be re-animated. Some say the novel can be classed as a romantic novel Margaret Drabble delineate romanticism as an extreme assertion of the self and The value of individual experience The stylistic keynote is intensity, and its Watchword is imagination. I think this suits to what we know about Mary Shelleys parents. I thought the novel is slightly complicated and takes a lot of time to understand the meanings behind it but overall a rise balanced, out of the ordinary but reasonably interesting novel. It shows what people were like in that time and straight away more a less within the first few chapters I was sympathising with the monster and my feelings towards the characters matched that of what Mary Shelley was trying to get across without blatantly saying it. Well it certainly aroused a endurance of scientists and philosophers when it was first publ ished and I think still to this day it makes people think and worry about these concepts becoming reality but not to the extremes of the riots and outrages they were keep going in the days it was published. It shows outrageous ideas in a more meaningful way and perhaps one day it might come true. Young adults like myself slightly naive to the laws of science, a book like this fuels the imagination and makes the impossible seem possible. Loved this book. Relevance Of Symbolic Play In Child Development Education Essay

Relevance Of Symbolic Play In Child Development Education EssayThis essay looks at the upshot of emblematical dawdle being remedial exemplify for nestlingren. The essay looks at the theory of typic get together, the use of symbolic play with baby birdren, the remedial value of symbolic play, using research to support the discussions. The essay discusses what is understood by symbolic play, then moves on to discuss the vastness of symbolic play in childrens development, tone at the motley theories that have been put forward to explain the importance of play in childrens development. The essay then moves on to look at conglomerate instances where therapeutic play has been used, and is considered useful, such as in children who are experiencing a chronic illness, who are hospitalized, who are awaiting surgery or who have go steadyd few form of trauma. What is symbolic play? Symbolic play is, according to Piaget (1962) one of the main ways in which children jibe to think in a representational manner, where representation is understood as a process through which children store information according to the perceptual analysis of their ingoing flummoxs (Leslie, 1987 Lyytinen et al., 1997). Play essentially allows children to study the things they experience and to put these things in to perspective. As such, play is fundamentally important for childrens development (Piaget, 1962). Play is one of the ways in which children understand the world, their interactions with others and the rules and regulations that govern their passage through the world. Play is, thus, a fundamental activity for children that allows them to form their personalities and to understand their place in the world, in relation to others.Relevant of symbolic play in child developmentSymbolic play, which arises around the age of 18 months, basically involves children substituting one item for a nonher, and git constitute incorporating others in to their play, pretend play or seque ncing. This can mean that children pretend to be mother (or father) or that they use household items to reproduce day-to-day activities they have seen their parents doing, amongst other common symbolic play activities. This symbolic allows children to experiment with different roles and different forms of being, which, as has been discussed, allows children to understand, and to contextualise, their experiences. As Piaget (1962) argues, such symbolic play allows children to understand the role of self and others, the use of objects and the sequences of actions and activities whilst playing. This allows children to understand objects, to learn about the properties of objects and to learn about consequences and causality (Piaget, 1962). Play thus allows children, amongst other things, to actualize where their boundaries are, to understand why things work in the ways they do and to understand how to interact with others. Symbolic play is thus, as has been discussed, a fundamental deve lopmental tool for children.Watson and Zlotlow (1999) talk about symbolic play as, an important characteristic of childrens azoic play, through the enactment of activities that are very familiar to the child in contexts that are not typical for those activities. Enacting activities out of context allows the child to seek, and to understand, the role of agents in their lives, allowing them to understand how the world works and what their role is in this world. Symbolic play, therefore, has been argued to have three main features the use of objects the use of actions during the play and individuals enacting the play (i.e., agents) (Watson and Zlotlow, 1999). The use of objects can be undertaken in umpteen forms, either through substitution, in which the object is used to represent mostthing else, through scenario-building, with the object being used to perform some task or in combination, with the object being used in various ways (Watson, 2008). The use of actions in play and the development of the agent in play follow certain patterns depending on the age of the child, as does the theme of childrens symbolic play (Watson, 2008). The one thing that all symbolic play has in common, at whatever age or developmental stage the child is at, is that the child uses some representation of self in their play. At age four, for example, children go out have progressed to fantasy play, entirely these fantasies will usually involve themselves in becoming their fantasy a fireman or a prince, for example (Garvey, 1990). This allows them to look different roles and responsibilities.Symbolic play is, therefore, a natural phenomena, that children follow naturally, as part of their development through childhood. As Reed (2007) argues, symbolic play colligate all four areas of a childs development, namely cognitive, language, social/emotional and physical, and, as such, is a fundamental part of the development of children. Symbolic play allows the child to contract knowle dge and to express and represent their ideas, thoughts and feelings as Vygotsky argues, in his sociocultural cognitive theory, symbolic play allows children to develop and to self-regulate and to work out any problems they might show (Reed, 2007). It is in this aspect, then, that the therapeutic value of symbolic play becomes clear. As children constantly experiment with their environment, their peers and the other agents they meet, and as the results of these experiments are constantly being fed screening to lead to reinforce certain behaviours or to elicit different patterns of behaviours, the value of play as therapy is clear. Children who are passing through a problematic second gear and who film assist to understand this, to contextualise these difficulties in the framework of their existing knowledge, can, through salty in therapeutic play, manage this. Therapeutic play allows them to experiment with the various outcomes and to contextualise the problems they vitrine, allowing them an opportunity to work through these problems/problematic situations, through this working out their difficulties and coming to understand how to deal with them. The use of symbolic play as therapyIn terms of the use of symbolic play as therapy with children, as Bettelheim states, Play permits the child to crock up in symbolic form unresolved problems of the past and to cope directly or symbolically with present concerns. It is also his most significant tool for preparing himself for the future and its tasks (Schaefer, 1995). Using play as a tool to help children through a difficult stage in their lives is, therefore, grounded in theory and makes sense in light of the fact that children learn through play. The research shows, for example, that therapeutic play can be helpful in aiding children through hospital stays (William, 2007) or through surgery, with this therapeutic play being shown to reduce solicitude and increase overall well-being (Bowmer, 2002). As Moore and Russ (2006) discuss, pretend play can act as a resource for children, relating, as it does, to many areas of adaptive operate including creativity, coping and emotional regulation. As Moore and Russ (2006) show, pretend play in hospital settings can reduce care and, through this, can reduce the likelihood of complications side by side(p) surgery and can allow children to adapt better to the complications of chronic illness. The therapeutic value of symbolic playIn terms of the therapeutic value of symbolic play, as Axline (1964) discusses, therapeutic play can be delivered through one of two major approaches, namely via non-directive play therapy and via directive play therapy (Oaklander, 1988). According to Axline (1964), play therapy should be governed by eight principles, namely that the therapist and child must be comfortable with each other that the therapist accepts the child as he/she is that the child can freely express their feelings that the therapist is alert to the feelings the child expresses that the therapist respects the child and their ability to resolve their own problems that the therapist does not attempt to influence the child that the therapy is carried out in its own time and that the only limitations placed on the play therapy are those that anchor the therapy in the real world (Axline, 1964). If the therapeutic play is provided within this framework, the benefits of the therapeutic play can be many and varied, as discussed above, with the troubled child benefitting greatly from this intervention. As Li and Lopez (2008) argue, therapeutic play can be valuable not only in helping children through a difficult stage in their lives but can be useful in helping to in prepare children for a stressful, or difficult, period that they might face. For example, children who are facing surgery or a chronic illness can be helped to come to terms with this through the use of therapeutic play. As Li and Lopez (2008) report, by reinforcing to nurs ing and medical examination staff that play is a very important part of the lives of young children, and be employing therapeutic play techniques, this can markedly improve the anxiety that these children experience and can allow these children to have a more positive outlook regarding their situation and the difficult things they will face, meaning that they are better equipped to face these situations. As Li and Lopez (2008) argue, therapeutic play can help to improve the resilience of children who face difficulties and, as such, it is an important tool to use in those children who need help to become more resilient.Children who have faced some form of trauma, for example, can be aided through the fallout of this trauma through engaging in therapeutic play. By allowing the child the space to act out what they saw, or what they have experient, for example, the child can come to terms with this traumatic experience. This therapeutic play could take the form of using puppets, or d rawing, or role playing anything that allows the child to re-enact the event(s) and, through this, come to terms with what happened. The use of therapeutic play in explaining the loss of someone nigh(a) to them is known to be particularly valuable, for example, as this can allow them to imagine the person is still here and to resolve any outstanding issues they had with them, or to slowly come to terms with the fact that they are no longer alive and no longer able to be physically present with the child. The simple act of playing can, therefore, allow the child to come to terms with many difficult emotions, allowing the child to slowly become more able to cope with the negative emotions that a traumatic event (such as witnessing a violent act or the unexpected death of a close family member), allowing them to deal with these negative emotions (Schaefer and OConnor, 1994). As Schaefer and OConnor (1994) argue, role playing in particular can help children to face the negative post-tr auma reactions that children might face, with role-play allowing children to relive the trauma and to insure various solutions to their negative experiences and emotions.ConclusionIn summary, then, this essay has looked at the issue of symbolic play being therapeutic play for children. The essay has looked, in particular, at the theory of symbolic play, the use of symbolic play with children and the therapeutic value of symbolic play. In regards to the theory of symbolic play, it has been seen that various theories have been proposed to explain the importance of symbolic play in childrens development, with all of these theories agreeing that play is a fundamental part of childrens development and that this play allows them to contextualise their experiences and to understand how these experiences dictate how they should act and should behave in different situations. It is this attribute of symbolic play that lends itself to being useful in helping children come to terms with difficu lt events in their lives as has been discussed, play allows children to explore different scenarios (even difficult scenarios), with this play enabling them to overcome any negative emotions or feelings they might have had towards these scenarios. A child who is facing a difficult surgery, for example, might be scared, might be worried for themselves and for their families, but engaging in role play with this child (for example) allows them the opportunity to explore different possibilities and to extend some of their anxieties and concerns. This has been shown, as discussed, to reduce the anxiety these children feel and to enable the childs health and well-being to be maintained, even under difficult circumstances. Similarly, a child who has experienced trauma can be helped through their post-traumatic period by engaging in some form of therapeutic play. This would normally consist of helping the child to act out their fears and their experiences, allowing the child to get rid of all of their negative emotions and feelings with regards to the even, allowing the child to expel these from their psyche and then to carry on without carrying this burden along with them.In conclusion, then, the essay has shown that play is fundamental for children and that, given its experimental nature and its ability to contextualise events and interactions for children, play is, at its very essence, therapeutic. Using play with children who need therapeutic interventions, then, makes perfect sense to allow children to understand the negative events/emotions and to overcome their fears and anxieties regarding these negative events and emotions. Therapeutic play has been shown to be a useful intervention in many cases, including in hospitalized children and in children living with chronic illnesses, who can be helped through this difficult period by engaging in therapeutic play, which has been shown to reduce their anxiety levels. In conclusion, then, therapeutic play is an exten sion of the symbolic play that children engage in as a normal developmental process and can be useful in helping children through difficult periods in their life.
